Nuclear Factor– B Induction by Visfatin in Human Vascular Endothelial Cells: Role in MMP-2/9 Production and Activation
نویسندگان
چکیده
Objective: Visfatin is elevated in obesity and T2DM; and thought to be an inflammatory mediator within atherosclerotic lesions inducing gelatinase activity. We investigated the activation of NFB a well-known pro-inflammatory transcription factor by visfatin in ECs. Research Design and Methods: Human ECs were transfected with pNFB-Luc plasmid. Using Quantitative PCR, western blot analysis and gelatin zymography, we studied NFB signalling in gelatinase mediated vascular inflammation by visfatin; employing the NF-kB inhibitor, BAY 11-7085. Results: Visfatin significantly increased NFB transcriptional activity (P<0.001). Also, we found a significant inhibition of TNFα induced NFB activity by visfatin (P<0.001). Furthermore, the NFB inhibitor, significantly negated visfatin induced MMP-2/9 mRNA expression, protein levels, and gelatinolytic activity (P<0.001). Conclusions: Visfatin induced NFB signalling in human ECs affects the activation of gelatinases-MMP-2/9, suggesting an important role of visfatin in the pathogenesis of vascular inflammation in obesity and T2DM.

Visfatin induces human endothelial VEGF and MMP-2/9 production via MAPK and PI3K/Akt signalling pathways: novel insights into visfatin-induced angiogenesis.
AIMS Visfatin is a novel adipokine whose plasma concentrations are altered in obesity and obesity-related disorders; these states are associated with an increased incidence of cardiovascular disease. We therefore investigated the effect of visfatin on vascular endothelial growth factor (VEGF) and matrix metalloproteinases (MMP-2, MMP-9) production and the potential signalling cascades. METHOD...

Chronic increases in arterial blood flow elicit an adaptive response of the arterial wall, leading to vessel enlargement and reduction in wall shear stress to physiological baseline value. Release of nitric oxide from endothelial cells exposed to excessive shear is a fundamental step in the remodeling process, and potentially triggers a cascade of events, including growth factor induction and m...
متن کاملVGB3 Induces Apoptosis by Inhibiting Phosphorylation of NF-κB p65 at Serine 536 in the Human Umbilical Vein Endothelial Cells
Background and objectives: Nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) inhibition results in an increase in apoptosis. It has been demonstrated that NF-κB subunit p65 phosphorylation at the IκB kinase phosphorylation site serine 536 (Ser536) is essential for the NF-κB nuclear translocation and activation. Therefore, NF-κB can be downregulated by suppressing its phosph...
متن کاملBlockade of monocyte-endothelial trafficking by transduced Tat-superoxide dismutase protein
It has previously been suggested that reactive oxygen species (ROS) are involved in the pathogenesis of chronic inflammatory diseases, which entails the initial activation of pro-inflammatory cytokines to facilitate leukocyte transmigration. The present study investigated whether intracellular superoxide dismutase (SOD) suppressed monocyte endothelial trafficking and transmigration. Human umbil...
متن کاملPatterns of Vascular Endothelial Growth Factor Expression in Hematopoietic Malignant Cells
Background and Objective: Vascular endothelial growth factor (VEGF) is a cytokine which is overexpressed in many malignant cancers including leukemia. VEGF plays an important role in tumor invasion and metastasis. Determination of the pattern of VEGF expression in human leukemic cell lines could be useful not only in screening of new antileukemic agents but also to study the mechanism of their ...
